The completed card must still be 2 1/2 by 3 1/2 inches and also must fit into the standard ATC plastic sleeve, which does give some flexibiity for thickness, but not a lot.
A typical tryptich can be 1/4 inch thick. I do have one that is just over 3/8 inch thick, but it still fits into its plastic sleeve. I am pretty flexible, but use some common sense. The post office charges more to mail anything over 1/4 inch thick, so it will cost you more to mail it to me, and it will cost me more to mail it back out with returns, so try to keep the total thickness 1/4 inch or less. :0)
